實(shí)驗(yàn)室介紹

尤忠勝教授分別在浙江大學(xué),中科院上海生化所、美國(guó)加州大學(xué)圣地亞哥分校(University of California, San Diego)生物系取得學(xué)士、碩士、博士學(xué)位,在索爾克生物研究所(Salk Institute)從事博士后研究。2009年成為美國(guó)圣路易斯華盛頓大學(xué)醫(yī)學(xué)院(Washington University in St. Louis , School of Medicine, WashU Medicine)細(xì)胞生物學(xué)與生理學(xué)系助理教授,2023年晉升為正教授。尤忠勝實(shí)驗(yàn)室主要研究細(xì)胞基因組維持、RNA質(zhì)量監(jiān)測(cè)以及先天免疫機(jī)制, 開(kāi)發(fā)針對(duì)腫瘤、自身免疫等疾病的新型治療策略和藥物。在Nature Medicine, Molecular Cell, Nature Cell Biology等刊物發(fā)表多篇高水平論文,報(bào)道原創(chuàng)性發(fā)現(xiàn)。本實(shí)驗(yàn)室為不同學(xué)術(shù)背景的研究者提供開(kāi)放的平臺(tái),誠(chéng)邀有志之士與我們共赴探索、創(chuàng)新之旅。尤忠勝教授具有豐富的學(xué)生和博后培養(yǎng)經(jīng)驗(yàn),曾任WashU分子細(xì)胞博士生培養(yǎng)項(xiàng)目(MCB Graduate Program)聯(lián)合主任、生物與生物醫(yī)學(xué)部(Division of Biology and Biomedical Sciences)執(zhí)行委員會(huì)委員。

1.全新基因組保護(hù)信號(hào)通路:發(fā)現(xiàn)并解析一條由胞質(zhì)DNA激活、 Ca2?介導(dǎo)的復(fù)制叉保護(hù)通路,鑒定了 10 余個(gè)核心調(diào)控因子(Li et al., Mol Cell 2019; Li et al., Mol Cell 2023; Kong et al., Nat Commun 2024)

2.TRPV2-STING互作: 首次發(fā)現(xiàn)STING與鈣離子通道蛋白TRPV2在內(nèi)質(zhì)網(wǎng)上的相互作用,該互作調(diào)控細(xì)胞內(nèi)鈣離子水平,并控制cGAS/STING先天免疫系統(tǒng)的靜息與激活(Li et al., Mol Cell 2023; Cheng et al., Cell Rep, accepted

3.無(wú)義 RNA 降解調(diào)控與腫瘤治療:構(gòu)建多個(gè) NMD 報(bào)告系統(tǒng),用于高通量藥物篩選與全基因組 CRISPR 遺傳篩選,鑒定多個(gè) NMD 抑制劑及調(diào)控因子;揭示 NMD 是攜帶剪接體突變的血液腫瘤的潛在治療靶點(diǎn) (Nickless et al., Nat Med 2014; Nickless et al., JBC 2017; Cheruiyot et al., Cancer Res 2021)

Laboratory Introduction

Dr. You is currently an Investigator at CIMR and Associate Director of the Chinese Institute for Cancer Research within CIMR. Prior to joining CIMR in 2025, he was a tenured professor at Washington University in St. Louis, School of Medicine. The You lab at CIMR investigates the molecular mechanisms of genome maintenance, RNA surveillance, and innate immunity, with the overarching goal of advancing cancer and autoimmune disease treatment. We integrate cell biology, structural biology, drug screening, and animal models to drive these studies.

Major Research Projects

The laboratory focuses on the following key research areas:

a.Intersection of DNA/RNA surveillance with innate immunity

b.Emerging roles of ions and ion channels in regulating cGAS/STING innate immune signaling

c.Therapeutic targeting of the TRPV2-STING axis in cancer and autoimmunity

d.Exploiting NMD vulnerability in spliceosome-mutant cancers

Major Contributions

1.Discovered a novel cytosolic DNA-elicited, Ca2+-mediated signaling pathway that safeguards DNA replication forks to preserve genome stability. (Mol Cell, 2019; Mol Cell 2023; Nat Commun, 2024)

2.Uncovered a novel interplay on the endoplasmic reticulum between the ion channel TRPV2 and the innate immune sensor STING, revealing a new mechanism for the co-regulation of Ca2+ dynamics and immune activation. (Mol Cell 2023; Cell Rep, accepted)

3.Pioneered the development of NMD reporter systems, enabling the identification of novel NMD inhibitors and genetic regulators, and demonstrated that NMD is modulated by intracellular Ca2+ and persistent DNA damage (Nat Med 2014; JBC 2017)

4.Established NMD as a promising therapeutic target for cancers harboring spliceosome mutations (Cancer Res 2021, Cancer Res, in review)

a. Bachelor's degree or higher in molecular biology, cell biology, biochemistry or related disciplines.

b. Proactive work attitude with strong organizational skills and attention to detail.
